摘要
从多粘菌素B处理的豚鼠腹腔渗出液中,分离嗜酸性粒细胞(Eos)用超声粉碎仪破坏细胞,离心后收集含Eos成分的上清液,给豚鼠经气管插管雾化喷入。60min后豚鼠对组胺的气道反应性PC20值显著降低(P<0.05)。Eos成分吸入处理组和正常对照组支气管肺泡灌洗液(BALF)中的细胞总数、Eos绝对计数差异均无显著意义(P>0.05)。但BALF细胞中上皮细胞的百分比显著增多(P<0.001),并且与60min时PC20值的降低呈显著正相关(r=0.84,P<0.05)。以上结果提示Eos可能通过其细胞成分造成气道上皮损伤诱导气道高反应性。
Eosinophils were collected from the peritoneal exudates of guinea-pigs pretreated with polymyxin B and the solution containing the protein of eosinophilic granules was prepared.After aerosolization,the solution was inhaled by guinea-pigs. It was found that the PC20 value was significantly decreased 60 minutes after inhalation(P<0.01).No obvious change of total cell count and eosinophil count but marked increase of epithelial cells in the bronchoalveolar lavage fluid(BALF)were found after inhalation,In addition,the increase of epithelial cells in BALF was correlated to the decrease of PC20.On the basis of these findings,it is concluded that eosinophils can induce hyperresponsiveness of the respiratory tract through the injury of its epithelium.
