诱导免疫耐受控制垂体移植免疫排斥Ⅰ.去脑垂体大鼠免疫耐受性的诱导

摘要 在立体定各仪经耳去除交系SD大鼠脑垂体基础上,用不同剂量近交新生的Wistar大鼠制成活化脾细胞和环磷酰胺进行诱导免疫耐受性的研究。结果:活化脾细胞5×10^7/0.5ml/W或5×10&6/0.5ml/W联合环磷酰胺20mg/kg/d持续2周后,去脑垂体大鼠同种异体皮肤移植自下而上时间分别为MST〉33.5天和MST〉47.8天。 Inbred spraque-Dawly female rats were hypophysectomized through the transeardrum stereotactic approach.Donor spleen Cells(SC)were isolated from inbred newborn wistar rats and activated by ConA.We induced the donor-specfic tolerance with the following processes,i.e.(1) injectong several doses of activated Spleen cells(ASC)onto the recipent(Hpox-SD rats)intravenously. (2)Cyclophosphamide(CP)intraperitoneally.(3)Combining ASC and CP.The result showed that ASC or ASC plus CP can promote skin allograft survial(MST>33.5 days and MST>56 days).The above results lead us to think that ASC or ASC plus CP can successfully induce donor-specfic tolerance in hypophysectomy rats.
