摘要
应用放射免疫分析疗法,研究了槲皮素对体外培养的人脐静脉内皮细胞释放内皮素(ET)的影响。结果表明:凝血酶和活化血小板分别与内皮细胞共同孵育4h和24h后,内皮细胞培养液中ET量显著增加(P<0.01)。槲皮素0.5、5和5μmol/L与内皮细胞预孵10min,可明显抑制凝血酶和活化血小板引起的内皮细胞大量释放ET。同时,槲皮素0.5~50μmol/L也可降低内皮细胞对ET的基础释放量。提示:槲皮素可抑制不同状态下的内皮细胞释放ET。
We performed a study to assess the effect of quercetin on release of endothelin by radioimmunoassay. Cultured endothelial cells (EC) isolated from human umbilical vein were used. Addition of thrombin or thrombin-activated platelets, resulted in increase of endothelin in the EC culture medium (P<0.01). Pretreatment of EC with quercetin at an amount of 0.5, 5 and 50 μM, the release of endothelin by EC stimulated with thrombin and activated platelets was potently inhibited. Furthermore, quercetin (0.5—50μM) also decreased basal production of ET from cultured human endothellal cells.
