L-甲状腺素对血管变态反应性收缩的影响及胞内钙调节机制

摘要 目的:考察正常豚鼠血管变态反应在L-甲状腺素给药后的病理模型中的改变。方法:制作L-甲状腺素致豚鼠血管损伤病理模型,作为甲亢组。以卵白蛋白作为抗原,诱发豚鼠离体血管变态反应。观察在有钙和无钙介质中及药物干预下,正常组和甲亢组血管变态反应性收缩的变化及对胞外和胞内钙的依赖性。结果:两组血管变态反应主要依赖于胞内钙,胞外钙的影响次之。胞内钙的作用主要涉及咖啡因敏感型钙池,去氧肾上腺素敏感型钙池的影响次之。结论:甲亢组发生血管变态反应时,胞内钙释放增加比正常组更明显,主要加强了胞内咖啡因敏感型钙池的钙释放。 Objective:To investigate the changes of allergic vascular response in L-thyroxin treated guinea pig pathological model. Methods: L-thyroxin treated guinea pig pathological model was prepared as hyperthyroidism group. The allergic vascular constriction was induced by ovalbumin in vitro. In calcium-contained or -free medium and under drug's interference, the changes of allergic vascular constrictions in normal and hyperthyroidism groups and the dependence on either extracellular or intracellular calcium was observed. Results: The allergic vascular constrictions in two groups were mainly dependent on the intracellular calcium release and less on the extracellular calcium. The effect of the caffeine-sensitive intraceUular calcium pool was most important, and the effect of the phenylephrine-sensitive intracellular calcium pool was less significant. Conclusion: An increase of the intracellular calcium release in hyperthyroidism group was more remarkable than that in normal group, which resulted mainly from increased intracellular calcium release from caffeine-sensitive calcium pool.
