鼻咽癌患者外周血树突状细胞体外扩增及生物学特性的研究 被引量:4

A Study of biological characteristic and generation of monocyte-derived dendritic cell from peripheral blood of patients with nasopharyngeal carcinoma

摘要 目的:观察鼻咽癌(NPC)患者外周血单核细胞来源的树突状细胞(DC)的形态、表型和功能与健康者DC的差别。方法:从12例NPC患者(NPC组)和9例健康者(对照组)的外周血中分离和培养DC,观察DC的形态,用流式细胞仪检测DC的表型CD83、CD1a、CD86和HLA-DR,用4,二甲基噻唑2,5二苯基四唑溴化物(MTT)比色法检测DC诱导混合淋巴细胞反应(MLR)的能力。结果:对照组的DC形态较NPC组成熟,NPC组CD83、CD1a的表达率较对照组低,差异有统计学意义(均P<0.05),但CD86、HLA-DR表达率2组差异无统计学意义(均P>0.05);NPC组CD83、CD1a、CD86和HLA-DR表达的平均荧光指数(MFI)均低于对照组,但差异无统计学意义(均P>0.05);对照组和NPC组外周血单核细胞均能诱导出DC,前者有较强的MLR诱导能力(P<0.05),而后者能力低下(P>0.05)。结论:NPC患者外周静脉血单核细胞来源的DC转化率较健康者低,且分化成熟能力也较低。 Objective:To study the differences of morphology,phenotype and function of dendritic cell(DC) derived from the healthy individuals and the patients with nasopharyngeal carcinoma (NPC). Method:DC were isolated and developed from peripheral blood monocytes from 12 NPC patients and 9 healthy volunteers in vitro, and were studied in terms of shape and phenotype of CD83,CD1a,CD86 , HLA-DR by flow cytometry then. MTT method was used to measured the capability of DC in mixed lymphocyte reactions(MLR). Result:The shape of DC derived from monocyte in the healthy volunteers peripheral blood was more typical than that of NPC patients'. The expression rate of CD83 ,CDla on DC from healthy volunteers was significantly higher than that from NPC pa- tients ( P 〈0.05). The DC from healthy volunteers had the higher expression level of CDla and CD83 than that from NPC patients, but not significantly( P〉0.05). DC in healthy volunteer's group could trigger proliferation of allogenous lymphocyte( P 〈0.05)but it could not significantly do that in NPC patient's group ( P 〉0.05). Con- clusion:These results reveal that the yield of DC derived from NPC patient's monocyte is lower than that from healthy individual's in peripheral blood,and the DC from NPC patient may be in an immature state and could not trigger the proliferation of lymphocyte.
