摘要 以苹果渣皮为原料,筛选到能在固态培养基中旺盛生长的丝孢酵母菌,研究了固态发酵的较佳条件:以苹果渣为主原料,丝孢酵母的适宜生长pH为6.0~6.5,最适生长温度为35℃,以尿素作氮源较好,培养基含水量在50%左右。在上述条件下,丝孢酵母ST851生长12h培养基pH稍有下降,24h降至6。菌体在24h内生长迅速。在此期间,可溶性糖的消耗速率与菌体蛋白的合成成正比;可溶性糖耗尽以后,半纤维素酶开始产生。一般发酵周期为24~36h,产品收率为87%。 Trichos poran sp.ST851 was selected which can grow well in solid medium using apple pomace ascarbon source.The suitable conditions of solid-state fermentation was studied.The optimal growing pH value and temperature were 6.0~6.5 and 35℃ respectively.Urea was a better nitrogen source.The water content of culture medium was about 50%.Under these conditions,pH value of culture medium reduced slightly in 12 hours after inoculation and dropped to 6 in 24 hours.During this course the consuming rate of soluble sugar was the direct ratio with the synthesis of cell protein.Hemicellulase was not synthesized until all soluble sugar was used out.The fermentation period was usually 24~36 hours,product yield was about 87%.
