摘要
在发现空弯菌43KD外膜通道蛋白不但是HSP60家族成员和优势抗原而且能诱导小鼠生产自身免疫应答的基础上,本文近一步分析了430KD优势抗原诱导产生针对ds-DNA、ss-DNA、ENA、组蛋白4种自身抗体的类型和意义。结果表明1.被诱导的抗ds-DNA、ss-DNA抗体主要是1gG,其次才是lgM型,偶见1gA型。2.被诱导的抗DNA、组蛋白自身抗体主要是IgM型和IgG型。3.IgM类抗DNA自身抗体和IgG类同时出现,这和前者产生细胞可能是后者产生细胞的前体这样一种推论矛盾。4.IgG、IgM抗ds-DNA抗体高峰和阳性率导以抗98-DNA抗体,提示正常B细胞库中对ss-DNA的免疫耐受更完全,识别ds-DNA的B细胞克隆数量多和/或轻易被选择活化。5.本文表明43KD优势抗原是空弯菌感染诱导自身免疫综合征小鼠模型中的关键性致病分子之一,为感染因子HSP诱导自身免疫病的理论假设提供了一种直接的实验证据。
In these years,we have successfully established a muine model with systemic autoimmune syndromes induced by chronic infection or immunization of Campylobacter jejuni,a common pathogen being infectious both in human and animals,and have found that 43 KD protein(HSP43)of C.jejuni was not only the major component of outer membrane proteins and immunodominant antigen but also a member of HSP60 family.In this paper we studied isotype of autoantibodies induced by by 43kDa immunodominant antigen of C jejun and got some results:1.isotype of autoantibodies against ds-DNA and ss-DNA was mainly IgG and then IgM;2.isotype of autoantibodies against ENA and histones was mainly IgM and then IgG;3 IgM and IgG against DNA were induced simultaneously,which was contrary to the inference that IgM might represent the precursor of pathogenic IgG to DNA;4.peak and positive percentage between IgG or IgM against ds-DNA and ss-DNA were different,suggesting that B cells tolerance to ss-DNA in primary B cells repertoire might be more complete than tods-DNA; 5.HSP43 was the key molecule of C jejuni-induced murine model with systemic autoimmune syndromes.Our results supported the hypothesis that immune response to HSP may be highly cross-reactive and even involve autoimmune diseases.
