Genetic Analysis and QTLs Tagging of Lint Percentage and Its Closely Related Yield Components in Upland Cotton

摘要 利用中国长江流域大面积种植的2个高衣分、高皮棉产量品种泗棉3号和苏棉16号,构建F2和F2∶3作图群体,进行高衣分及铃重、单铃种子数、籽指、衣指和百粒籽棉重等相关性状的QTL定位和分子标记筛选。利用P1、P2、F1、F2和F2∶3等群体进行联合世代遗传模型分析。结果表明:衣分以及上述相关性状多符合主基因+多基因混合遗传模型;用2 131对SSR和1 040个RAPD引物进行双亲多态性检测,仅37对SSR和5个RAPD引物具有多态性,产生43个稳定的多态性位点;QTL作图检测到的2个衣分的QTL,在F2和F2∶3群体中都能检测到。另外还检测到控制衣指的1个QTL和控制百粒籽棉重的3个QTL,与这些QTL紧密连锁的分子标记可以用于对衣分及其相关性状的分子标记辅助选择。 Simian 3 and Sumian 16 had been two cotton cultivars grown extensively in the Yangtze valley with high yield and high lint percentage. Lint percentage of Sumian 16 is higher than that of Simian 3. The method of major gene plus polygene mixed inheritance model was used to analysis the genetics of lint percentage and its closely related yield components using jointly segregation analysis of multiple generation. Most of the optimum models of the lint percentage and its closely related yield components are major gene plus polygene mixed inheritance model. QTL tagging of lint percentage and its closely related yield components in (Sumian 16 × Simian 3 )F2 and F2:3 generation were conducted using 37 SSR and 5 RAPD markers having polymorphism between parents screened from a total of 2 131 pair SSR primers and 1 040 RAPD primers. The results showed that 2 QTLs controlling lint percentage had been tagged in both F2 and F2:3 based on composite interval mapping and marker multiple regression methods as well as single-marker analysis method. Simultaneously, 1 QTL controlling lint index and 3 QTLs controlling seed cotton weight of 100 seeds had been screened in F2 and F2:3 generation. The molecular markers tightly linked with the QTLs controlling high lint percentage and its closely related yield components can be used in MAS for high lint percentage.
