摘要 目的:观察末端病大鼠跟腱末端区内皮型一氧化氮合酶(eNOS)的分布,探讨eNOS与血管新生的关系及其在末端病中对机体的保护作用。方法:将8只Wistar大鼠随机分为对照组和造模组,采用电刺激跳跃法造模,4周后处死动物,取其双后足跟腱及部分跟骨,制成观察标本,采用ImageProPlus5.0图像分析系统测定标本镜下视野照片的阳性信号积分光密度值(IOD)。结果:对照组主要在腱围处有eNOS的大量表达;造模组大鼠跟腱、纤维软骨、钙化软骨、跟骨、骨髓腔、腱骨关节面及腱围等部位eNOS表达明显,腱围及骨髓腔处表达最多。两组间不同部位IOD值均有显著差异。结果提示:eNOS可能参与了末端病组织血管的新生,组织结构功能适应及抑制细胞凋亡等代偿性反应。 Objective This study was to investigate the distribution of nitric oxide and the protective effect of endothelial nitric oxide synthase (eNOS), as well as the relation of eNOS to angiogenesis in enthesiopathy. Methods Eight Wistar male rats (weight 200g) were randomly divided into the control group ( n = 2) and the experimental group ( n = 6). Rats in experimental group were forced to jump upwards through electrical stimulation for 4 weeks, then all rats were killed, and calcaneal tendon and part of calcaneal bone were collected for preparing the slides. The expression of eNOS in calcaneal tendon entheses were detected and analyzed by Image- Pro Plus 5.0 Image- Analysis System. Results There were strong expression of eNOS in peritendon in control group, and the expression of eNOS in tenodn, unmineralized fibrocartilage, mineralized cartilage, calcaneus, cavitas medullaris, and facies articularis ossium in the experimental group were significantly stronger than that in control group. The values of IOD in various parts between two groups were significantly different. Conclusion The results suggested that the eNOS involved in the angiogenesis, the improvement of tissue rigidity and the inhibition of apoptosis in enthesiopathy.
