摘要
选用C18预处理柱分离出23例慢性充血性心力衰竭(CHF)患者血浆内源性类洋地黄物质(EDLS),应用高压液相色谱(HPLC)对其进行部分纯化,观察部分纯化的EDLS对红细胞摄取86Rb的抑制,而对其进行鉴定,比较部分纯化的EDLS与哇巴因在C18反相分析柱上的保留时间。结果表明:①CHF患者血浆第二份HPLC洗脱物(保留时间3~6分钟)对红细胞摄取86Rb具有强烈的抑制作用;②其抑制率为80%±6%,明显高于其他洗脱物份别及健康对照血浆第二份洗脱物的抑制率(43%±3%)(均P<0.01);③部分纯化的EDLS、哇巴因及两者混合物的保留时间基本一致,提示EDLS与哇巴因结构相似。
The endogenous digitalis-like substance(EDLS) was isolated by C18 disposable columns and partially purified by high performance liquid chromotography(HPLC) in the plasma of 23 patients with CHF and 12 healthy controls. Inhibition of partially purified EDLS on 86Rb uptake by intact erythrocytes was observed to determinate the EDLS,The chromatographic retention of partially purified EDLS on the C18 reversed phase column by HPLC was compared with Ouabain.The results showed:①Fraction Ⅱ in the plasma of patients with CHF possessed stronge inhibitive eftect of 86Rb uptake by intact erythrocytes;②The inhibition rate was 80%±8%,which was obviously higher than the other fractions in the same sample and the fraction Ⅱ of normal controls(43%±3%) (all P<0.01).③The retentions of EDLS. Ouabain and the mixture of both were almost equal. It suggests that EDLS may have same structure as Ouabain.
