摘要
本实验以生理盐水、iloprost及其改良液为保存液,在4℃保存兔胎脑皮层组织,以高效液相色谱分析仪(HPLC)测定在保存过程中的腺苷酸含量变化,以了解胎脑组织在4℃保存时的能量代谢情况。结果显示,在生理盐水组及iloprost液组,随保存时间延长,胎脑组织中的三磷酸腺苷(ATATP)含量、能量负荷(EC)水平明显下降(P<0.01),在保存2小时接近耗竭,且两组间无明显差异(P>0.05);而改良iloprost液组的ATP含量及EC水平在保存2小时却略高于保存1小时(P>0.05)。此结果表明,要改进4℃保存的效果,不仅需应用细胞保护剂,还应考虑如何延缓能量衰竭的发生。本实验通过在iloprost液中加入ATP等物质,获得了较iloprost液更好的保存效果。
e researcbed the adenine nucleotide contents of the embryonic cortical brain tissue of rabbits withHPLC, which were preserved in saline, iloprost solution (50ng/ml) and improved iloprost solution at 4℃,in order to colnprehend the energy metabolism of preserved embryonic cortical brain tissue. The resultsindicated: the content of adenosine triphosphate (ATP) and energy charge (EC) level of preserved tissuein saline and iloprost solution clearly declined as the preservation time prolonged (P<0.01), and tendedto exhaustion at 2 hours preservation, there was not significant distinction between these two groups; at2 hours preservation, the content of ATP and the level of EC of the preserved tissue in improved iloprostsolution were less higher than at 1 hour preservation. So, in order to improve the effect of preservationat 4℃, it is necessary that the cellular protectant is used, but how to retard the progress of energyexhaustion is considered as well. In this study, we obtained the better effect of preservation with improvediloprost solution than iloprost solution, it is because there axe ATP and other substance in the improvediloprost Solution.
