摘要
目的:研究冠心病患者外周血淋巴细胞低密度脂蛋白(LDL)受体信使核糖核酸(mRNA)的表达。方法:用反转录聚合酶链反应(RT-PCR)法对人LDL受体互补脱氧核糖核酸(cDNA)片段进行克隆。由此得到的质粒脱氧核糖核酸(DNA)在克隆片段特定位点切开,插入小的DNA片段行再连接。此质粒DNA在体外转录成mRNA,其在定量PCR中作为一个内标准。用此法定量测定了15例冠心病及15例对照组患者外周血淋巴细胞LDL受体mRNA。数据处理用t检验。结果:冠心病患者血淋巴细胞LDL受体mRNA水平明显低于对照组(P<0.001)。结论:结果提示冠心病患者存在LDL受体mRNA表达不足。
Objective:To investigate LDL-receptor mRNA expression in patients with CHD. Methods:With reverse transcription-polymerase chain reaction (RT-PCR),the LDL-receptor complementary DNA has been subcloned for in vitro transcription. The resutling plasmid DNA has been cut at a unique restriction site of the cDNA sequence and religated after inserting certain small DNA fragments into the same site. After in vitro transcription, this allelic variant mRNA was used as an internal standard in PCR. The levels of LDL receptor mRNA of peripheral blood lymphocytes in 15 patients with CHD and 15 controls had been evaluated by quantitative PCR. The difference between groups was analyzed by t test. Results: The level of LDL receptor mRNA of the CHD patients was significantly lower than that of controls(p<0.001). Conclusion: The findings suggest that the expression of LDL receptor mRNA is insufficient in patients with CHD.
