牛卵泡颗粒细胞无血清培养的形态学观察 被引量:3

摘要 以M cCoy′s5a为基础培养液添加硒、转铁蛋白、雄烯二酮、谷氨酰胺,对牛卵巢上大(直径>8 mm)、中(4 mm<直径<8 mm)、小(直径<4 mm)非闭锁卵泡的颗粒细胞进行无血清培养。结果表明,M cCoy′s5a无血清培养中的颗粒细胞形态与分化特点类似于体内情况,说明颗粒细胞在该培养条件下,其功能可能更接近体内的生理状况。 A new culture in vitro method for bovine granulosa cells from non -atretic follicles at different size(small:〈4 mm,medium:4-8 mm,and largt: 〉8 mm diameter) and a serum-frue culture system using McCoy's 5a medium(pH7.4) supple-mented with 20 mmol/L Hepes,3 mmol/L L-glutamine.0.1% BSA .29 μg/L, testosterone.2. 5 mg/L transterrin and 4μg/L setenium were developed, The results indicated that morphological characteristics of granulosn cells was similar to that of in vivo in this long-term culture system.
