摘要
目的:观察兔不同程度面神经损伤后,损伤局部与创伤愈合和神经再生相关的转化生长因子β1表达的变化。方法:实验于2004-06-09/2005-02-30在解放军第四军医大学西京医院实验外科完成。取成年长耳白毛家兔68只,随机分为6组,对照组(n=9) 和5个损伤组,损伤组分别在双侧面神经颊面干造成暴露(n=13)、牵拉 (n=10)、压榨(n=11)、挤压(n=12)及切断(n=13)5种损伤,于伤后1 h、 1,3,7 d分别取材。苏木精-伊红染色行组织学观察;转化生长因子β1 免疫组化染色,按照细胞着色的程度行染色值半定量分析。结果:68只兔进入结果分析。对照组转化生长因子β1染色值为1.27±0.31, 暴露组、牵拉组各时间点转化生长因子β1表达无明显变化,压榨组、挤压组、切断组面神经损伤后1 h和1 d转化生长因子β1表达升高(P<0.05), 3 d时下降,7 d时再度出现升高,显著高于对照组(1.99±0.32,2.04±0.29. 2.56±0.42,P<0.05)。各组间相比可见,同一时间点不同损伤组的染色值为切断组>挤压组>压榨组>牵拉组>暴露组。结论:①不同程度面神经损伤后,损伤局部的转化生长因子β1水平随损伤程度加重而增高。②损伤后转化生长因子β1水平在1 d和7 d各出现1个高峰。提示转化生长因子β1的表达在面神经损伤后的病理变化与修复再生过程中具有重要意义。
AIM: To observe the change of local transforming growth factor beta (TGF-β1) expression, which is correlated with wound healing and neural regeneration in rabbits after facial nerve injury of different degree. METHODS: The experiment was carried out in the laboratory surgery department of Xijing Hospital, the Fourth Military Medical University of Chinese PLA between June 2004 and February 2005. Sixty-eight adult rabbits were randomly divided into 6 groups: control group (n=9) and 5 injury groups, rats in the injury groups were made into bilateral facial nerve injury models of exposure (n=13), stretch (n=10), compression (n=11), crush (n=12) and cut (n=13). The materials were taken at 1 hour and 1, 3 and 7 days after injury. The hematoxylin-eosin (HE) staining was applied in the pathological observation. The TGF-β1 was detected with immunohistochemical staining, and the semi-quantitative analysis of the stained value was performed according to the degree of cell pigmenting. RESULTS: All the 68 rabbits were involved in the analysis of results. The stained value of TGF-β1in the control group was 1.27±0.31, the expression of TGF-β1 in the exposure group and stretch group had no obvious changes at different time point. In the compression group, crush group and cut group, the expressions of TGF-β1 were increased at 1 hour and 1 day after the facial nerve injury (P〈 0.05), decreased at 3 days, and increased again at 7 days, which were all significantly higher than that in the control group (1.99±0.32, 2.04±0.29, 2.56±0.42, P〈 0.05). At the same time point, the stained value was that cut group〉 crush group〉 compression group〉 c ompression group〉 exposure group. CONCLUSION: ①After facial nerve injury of different degree, the local TGF-β1level is increased with the aggravation of injured severity. ② The TGF-β1 level had a peak at 1 and 7 days after injury respectively, indicating that TGF-β1 expression plays an important role in the pathological changes and recovery after facial nerve injury.
