摘要
采用CTAB法提取金花梨基因组DNA,在磨样时,1 g鲜样添加0.1 g Vc,提取的DNA产率较高,且极易溶解,能满足RAPO及其他分子生物学研究的需要。试验还发现,最佳提取材料是嫩芽,其次是新梢和成熟的叶片。
Genomic DNA of Jinhua Pear was extracted by CTAB metheds. Results showed that with the dose of 0.1. g/g of Vc added when the samples were triturated, genomic DNA yield was high and it was easiest to dissolve. And it was optimal for RAPD analysis and other molecular biology researches. The results of the experiment also showed that tender bud was the best material, and the next ones were shoot and mature leaf.
