摘要
以玉米为原料,研究了超氧化物歧化酶(SOD)的提取及对酒精发酵的影响。玉米SOD提取的最佳工艺条件为:100g玉米加入0.05mol/L磷酸缓冲液200mL,40℃浸泡36h;淋干后加入0.05mol/L磷酸缓冲液200mL,破碎浸提1h,离心得SOD提取液,活力为38078U;然后添加硫酸铵至饱和浓度90%进行盐析,SOD冻干粉的比活为168.3U/mg(蛋白)。而用提取SOD后的玉米渣进行酒精发酵,其淀粉出酒率达到54.25%。另外,还分别考察了玉米浸泡液和SOD盐析液对酒精发酵的影响。
The extraction of superoxide dismutase from maize and its effect on ethanol fermentation were studied. The optimal extracting conditions of SOD were that 200 mL. 0.05 mol/L phosphate buffer at 40℃ for 36h; then the smashed maize was extracted with 0.05 mol/L phosphate buffer 200 mL for lh, the extracts containing SOD were obtained by centrifugation, whose total SOD activities were 38078U. Ammonium sulfate was added with stirring to SOD extracts to make a 90% saturated solution, and SOD powder was gained by lyophilization, whose specific activities were 168.3 U/rag protein. Moreover, the ethanol yield of starch reached 54. 2596 when ethanol fermentation was processed with maize residue after extracting SOD. The effects of dipping liquid and the remained salt out solution on ethanol fermentation were discussed.
