反相高效液相色谱法检测酵母胞内V_B_1含量 被引量:3

Determination of Intracellular Thiamine of Yeast by RP-HPLC

摘要 采用反相高效液相色谱法测定了酵母胞内VB1的含量。采用ODS2HYPERSIL色谱柱(5μm,250×4.6mm),优化后的色谱条件为:流动相V(甲醇)∶V(水)=3∶7,流速1.2mL/min,波长238nm,柱温35℃。在此条件下回收率为88%~92%。此方法步骤简单,重现性好。 A quantitative method was developed for the determination of Vitamin B1 in the yeast cells by RPHPLC. The chromatographic column was ODS-2 HYPERSIL (5 μm,250×4.6 mm) .The optimal chromatographic conditions including methanol and water at 3:7 were served as mobile phase, and flow rate was 1.2 mL/min at 238 nm by using UV-detector; column temperature was 35℃. The methods recovery rate was 88 % 92%. The method is simple and reproducible.
