摘要
通过RT-PCR技术从橡胶树中克隆了CBF基因家族的保守区,并通过5'端RACE和3'端RACE技术克隆了全长为1121bp的cDNA序列,通过BLAST工具搜索Genbank数据库。结果表明,该cDNA序列同其他植物中的CBF家族基因高度同源,认为该cDNA序列就是橡胶树中的CBF基因。利用genescan和ClastalX工具对该序列进行分析,推测其编码区为693bp,编码231Aa,并包含一个AP2DNA结合域。氨基酸同源性分析结果表明,来自橡胶树的CBF基因氨基酸序列与热带起源的作物同源性较高,而与北方种植作物相似性较低。
The conserved domain of CBF gene in Hevea brasiliensis was cloned, and the 5' terminal and 3' terminal sequences were amplified by using RACE technique. By assembling all the sequences, a contig of 1 121 basepairs was obtained. The results of searching the Genbank by blast methods showed that this sequence was highly homologous with the CBF genes in the other plants, and was deduced as the CBF gene in the rubber tree. The sequence was analyzed by the genescan tool, which was deducted to be an ORF of 693 basepairs. The ORF was encoded a polypeptide sequence of 231 amino acids, in which an AP2 DNA binding domain was found. Homology analysis was carried out with the ClastalX, and the results showed that the HbCBF1 gene had higher homology to the CBF genes of tropical crops than to those identified in the crops planted in the temperate zone.
