摘要
目的:研究复康片预防卡马西平(CBZ)肝毒性的效果。方法:将48只大鼠随机分为正常对照组、模型对照组、复康片低剂量组、复康片高剂量组共4组,每组12只。正常对照组正常喂养,每d灌胃蒸馏水,其余不做任何处理。余3组建立CBZ致消化系统损伤模型,造模同时复康片低、高剂量组分别按750mg/(kg·d)和1500mg/(kg·d)灌胃复康片1次/d。90d后,脊髓脱臼处死大鼠,腔静脉取血,离心取血清测肝功能,包括天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、碱性磷酸酶(ALP),白蛋白(ALB),取肝组织在光镜与电镜下观察。结果:与正常对照组比较,模型对照组肝功能异常,线粒体面积、脂肪滴面积增大,线粒体密度降低,肝脏病理为受损表现。与模型对照组比较,复康片高剂量组大鼠血清AST、ALT、ALP降低,ALB升高;肝细胞核膜结构清晰完整,核周无水肿,粗面内质网结构正常,含有丰富的核糖体,线粒体形态结构正常,线粒体面积与密度、脂肪滴面积恢复(P<0.05)。复康片低剂量组ALT降低,ALB升高;肝细胞线粒体基本正常或偶有轻度水肿,脂肪滴面积较模型对照组减少(P<0.05)。结论:复康片对CBZ所致大鼠肝功能、肝脏超微结构损害有预防性保护作用,以高剂量为优。
Aim: To study the preventive effect of Fukang tablet on liver toxicity induced by carbamazepine(CBZ) . Methods: A total of 48 rats were randomly allocated into 4 groups:normal control group was given distilled water, model group,low-dose and high-dose Fukang tablet groups were given CBZ to establish the digestive system injury model. At the same time,low-dose and high-dose Fukang tablet groups were given Fukang tablet at 750 mg/kg and 1 500 mg/kg per day by gavage, respectively. After 90 d, rats in 4 groups were executed and blood samples were taken for evaluating the liver function with the following parameters:AST,ALT,ALP, and ALB. The liver tissue was also observed under light microscope and electron microscope. Results: Compared with normal control group, the liver function was abnormal,the areas of mitochondrion and fat drop enlarged,the density of mitochondrion decreased and the liver was injured pathologically in model group. Compared with the model group,the 2 Fukang tablet groups had lower levels of AST, ALT,and ALP and higher level of ALB of hepatocytes. Conclusion: Fukang tablets,especially with high dosage, can protect the rats from the liver toxicity of CBZ.
