RNAi对人胃癌BGC823细胞β-catenin基因的抑制作用

摘要 目的:应用RNAi技术抑制胃癌BGC823细胞βcatenin基因的表达,探讨该基因在胃癌细胞生长增殖中的作用。方法:体外转录合成短发夹状RNA,用脂质体转染系统将其导入细胞内。应用RTPCR检测转染后目的基因mRNA的表达水平;应用MTT检测转染后癌细胞生长增殖情况。结果:RTPCR电泳结果显示RNAi可抑制胃癌细胞βcatenin的表达,第2d和第3d抑制率可达90%以上。MTT实验显示βcatenin基因受抑后细胞的生长增殖受到明显抑制,抑制率最高达43%;且抑制率随着时间而升高。结论:RNA干扰技术作为一种新的基因治疗技术在胃癌治疗方面有着巨大的应用潜力。 Aim: To investigate the inhibitive effects of RNA interference(RNAi) on the expression of β-catenin in gastric cancer BGC-823 cells. Methods: The DNA template and T7 RNA polymerase were used to synthesize the short hair- pin RNA (shRNA) , which was transfected into the BGC-823 cells by Transfection Reagent. RT-PCR was used to detect the expression of β-catenin. MTT was used to investigate the cellular proliferation after the transfection. Results: RT-PCR showed that β-catenin was inhibited by RNAi. The highest inhibitory rate was above 90%, The result of MTT indicated that when 13-catenin expression was inhibited, the cancer cellular proliferation was restrained remarkably,the highest inhibiton- grate was 43% and the inhibitory rate would increase with time. Conclusion: RNAi has great potential in the treatment of gastric cancer as a new gene therapy.
