人脑星形细胞瘤组织转化生长因子α及表皮生长因子受体检测 被引量:1

Detection of TGF-α and EGFR in human astrocytoma tissue

摘要 目的:研究转化生长因子α(TGFα)及表皮生长因子受体(EGFR)的表达与人脑星形细胞瘤发生发展的关系。方法:用原位杂交及免疫组化技术检测50例人脑星形细胞瘤(其中星形细胞瘤18例(WHOgradeⅠ~Ⅱ级),间变性星形细胞瘤20例(WHOgradeⅢ级),多形性胶质母细胞瘤12例(WHOgradeⅣ级))和10例正常脑组织中TGFαmRNA、TGFα蛋白及EGFR蛋白的表达。结果:正常脑组织中无TGFα和EGFR表达。50例星形细胞瘤TGFαmRNA阳性率为56%,TGFα蛋白和EGFR蛋白的阳性率分别为64%和68%,3者的表达水平均与肿瘤的病理分级呈正相关(P<0.01)。TGFα蛋白与EGFR蛋白表达均存在相关性(P<0.05)。结论:人脑星形细胞瘤有TGFα基因异常表达,TGFα/EGFR自分泌系统在肿瘤发生发展过程中起重要作用。 Aim : To study the expression of TGF-α gene and EGFR and their relationship to occurrence and development of human astrocytomas. Methods: In situ hybridization and immunohistochemistry were used to investigate the expression of TGF-α mRNA, TGF-α, EGFR in fifty human astrocytoma samples and ten cases of normal brain tissue, respectively. Results: There was no positive expression of TGF-α protein or EGFR protein in normal brain tissue. The positive staining rate of TGF-αmRNA in human astrocytoma tissue was 56% . The total positive rates of TGF-α and EGFR protein were 64% and 68% , respectively in human astrocytoma samples. The expressions of the 3 parameters were positively correlated with the pathological grades (P 〈0. 01 ). TGF-α was positively correlated with EGFR ( P 〈0. 05 ). Conclusion : It is suggested that there is abnormal expression of TGF-αgene in human astrocytoma tissue and the autosecretion system of TGF-α EGFR may play a key role in the oncogenesis and development of human astrocytomas.
