摘要
本研究以常规SDS-PAGE和电洗脱技术相结合,分离纯化旅毛虫肌肉期幼虫分泌排泄物(TSLI-ES)中46-58kD的蛋白组份,并用于Dot-ELISA诊断旋毛虫病.检测结果:旋毛虫病人血清34份,阳性32份,阳性反应率为94.12%;正常人血精100份未发现假阳性;对蛔虫病人(12份)、囊虫病人(15份)、血吸虫病人(15份)及肺吸虫病人(20份)血清亦无交叉叵应出现;建立的Dot-ELISA易于操作.重复性和稳定性较好。用Dot-ELISA比较纯化抗原和TsL1-ES的诊断效果,结果经卡方检验后显示:用纯化抗原和用TsL1-ES检测的敏感性无显著差异,但纯化抗原的特异性显著优于TsL1-ES。
The 46-58 kD proteins from excretory/secretory products of Trichinella spiralis muscle larvae(TslL1-ES) were seperated and purified by common SDS-PAGE and electroelutiontechnique, and applied for diagnosing trichinosis with Dot-ELISA. The results showedthat the positive rate was 94. 12% (32/34) in cases of trichinosis, but no false positive andcross reaction were found in sera from 100 normal individuals, 12 sera of ascariasis, 15sera of cysticercosis, 15 sera of schistosomiasis and 20 sera of paragonimiasis. The purifiedantigens were compared with that of TsL1-ES by Dot-ELISA. There was no significantdifference in sensitivity between the two antigens but in contrast, the specificity of purifiedantigens was significantly higher than that of TsL1-ES.
