reelin相关基因研究进展 被引量:9

摘要 在脑发育过程中,新产生的神经元由内向外从脑室附近向脑膜迁移,即新形成的神经元要越过先形成的神经元最后定位于皮层的外表面,由此形成了大脑皮层、小脑皮层和海马等特有的分层结构。当reelin、VLDLR、ApoE R2和Dab1基因突变时,细胞迁移及定位方向发生改变,各皮层的细胞构筑有明显缺陷,动物出现共济运动不良。信号传导研究发现reelin与受体VLDLR、ApoE R2和整合素结合后,促使Dab1磷酸化,启动信号传导系统,调节了细胞与细胞以及细胞与基质之间的黏附分子作用,与神经元的正确迁移以及大脑的正常发育有关。当此信号传导中任何一个元件发生错误便可能导致精神神经系统疾病。
