摘要
以高压静电法制备转BMP-2基因骨髓间充质干细胞的海藻酸钠-聚赖氨酸-海藻酸钠微囊,考察了细胞浓度、海藻酸钠浓度、细胞悬液离心次数对圆囊率、粒径分布和成囊性的影响。结果表明,细胞浓度3×106个/ml,海藻酸钠浓度1.75%,细胞悬液离心洗涤2次的条件下,能制得球形好、粒径为200~300μm的微囊。
Alginate-polylysine-alginate microcapsules loaded with recombinant BMP-2 gene bone marrow mesenchymal stem cells (MSCs) were prepared by high-voltage electrostatic method. The factors such as the concentration of MSCs and sodium alginate and the times of centrifugation affected on the integrity and diameter distribution of microcapsules were investigated. The results showed that the spherical microcapsules with 200 - 300μm diameter were obtained under the conditions of MSCs suspension of 3×10^6/ml and 1.75%sodium alginate, centrifugated for 2 times.
