摘要
目的从中药苦参中快速分离纯化氧化苦参碱。方法采用反相C18柱色谱与正相低压干柱柱色谱从苦参的醇提物中分离纯化氧化苦参碱。结果经优化操作条件的反相柱色谱可去除非极性和极性较小的成分,采用低压干柱柱色谱可快速、有效将氧化苦参碱与其他生物碱成分、极性化合物分离,获得的氧化苦参碱与标准品比较结构、性质一致。结论根据正相色谱和反相柱色谱分离特点,建立了分离纯化氧化苦参碱的方法。
OBJECTIVE To develop a method fi)r the separation and purification of oxymatrine with C18 reversal-phase chromatography and normal dry-cohumn flash chromatography. METHODS Oxymatrine was separated and purified with C18 reversed-phase chromatography and normal dry-cohumn tlash chromatography, respectively. RESULTS The operating conditions of the two ehromatographie steps were investigated and optimized. Reversed-phase chromatography removed non-polar and lower polarity compounds. The normal dry-eohtmn flash chromatography separated quickly for oxymatrine from alkaloid malrinc mixture and polar compounds. The strueture and character of oxymatfine were identified to the oxymatfine standard. CONCLUSION A new separation and purification system for oxymatrine was established with normal and reversed-phase chromatography.
