反相高效液相色谱测定地骨皮中莨菪亭的含量

Determination of scopoletin in Cortex Lycii by RP-HPLC

目的建立反相高效液相色谱测定地骨皮中莨菪亭含量的方法。方法使用DiamonsilC18色谱柱(4.6mm×200mm,5μm),流动相为甲醇-冰醋酸水溶液(1→100)(25:75,pH=3.0),检测波长为345nm。结果在0.0018-0.1440μg内莨菪亭呈现良好线性关系(r=0.9999)。RSD<1.3%。结论用本法测定地骨皮中莨菪亭含量灵敏准确,为该药材的质量评价提供了一定的参考依据。

OBJECTIVE develop a RP-HPLC method to determine the content of scopoletin in Cortex Lycii from different sources. METHODS The separation was accomplished on a Diamonsil C18 column （4.6 mm × 200 mm, 5μm） with a mixture of methanol- aqueous acetic acid （ 1→ 100） （25：75, pH = 3.0） as the mobile phase. The eluates were deteeted at 345 nm. RGSULTS The linear range was 0.001 8-0.144 0 μg （ r= 0.999 9）. The precision was perfect （RSD〈 1.3%） .CONCLUSION This study lind adequate sensitivity and accuracy to measure the contents of scopoletin and could be used in the quality control of Cortex Lycii.
