反相高效液相色谱法测定小鼠血清及脏器中氟尿苷的浓度

摘要 目的:建立以反相高效液相色谱法快速测定小鼠血清及部分脏器匀浆中氟尿苷浓度的方法.方法:色谱柱为C18,流动相为磷酸盐缓冲液(pH=7.4)-甲醇(95:5),检测波长为268nm.结果:氟尿苷检测浓度线性范围均为15~240μg/ml,最低检测浓度为3μ/ml(S/N≥3);平均提取回收率>95%,方法回收率在91%~112%之间,日内和日间精密度均<10%.结论:本方法简便、快速、可靠、经济,可用于氟尿苷血药浓度分析及动物体内分布研究. OBJECTIVE: To establish RP- HPLC method for the fast determination of floxuridinein in serum and homogenate of some of the organs of mice. METHODS: The determination was performed on C18 chromatographic column with its mobile phase consisted of phosphate buffer(pH = 7.4) - methanol(95 : 5) and with its detection wavelength at 268nm. RESULTS: Linear detection concentration ranges of floxuridinein were all at 15-240μg/ ml. The minimum detection concentration was 3μg/mL(S/N≥3) . The average extraction recovery was above 95% . The average recovery ranged from 91% to 112%. Both the intra-day and inter-day precisions were less than 10%. CONCLUSION: The method is simple, rapid, reliable and economical, and it can be used for the determination of serum floxuridinein concentration and the study of its body distribution in animals.
