摘要
目的:研究肠道神经系统中神经肽类激素促皮质激素释放激素(CRF)和P物质(SP)相关神经元的神经解剖关系及CRF对黏膜下神经丛SP分泌的影响。方法:采用免疫荧光组织化学染色及双染色研究CRF神经元与SP神经元的神经解剖关系;体外肠神经组织培养及放免法研究CRF对SP分泌的影响。结果:豚鼠黏膜下层神经丛中所有表达SP的神经元上均有CRF1受体的表达。CRF可促进体外培养神经组织分泌SP。结论:在肠道神经系统CRF及其受体与SP在神经解剖和生理功能方面均存在密切的关系,CRF通过与分泌SP神经元上的CRF1受体结合促进SP的分泌。
Objective To investigate the neuro-anatomical relationship between the neurons expressed eorticotropin releasing hormone (CRF)1 receptor and the neurons secreting substance P (SP) and the physiological function of CRF on SP secretion in submucosal plexus, Methods Immunofluorescene stain and double stain were adapted to study the neuro-anatomical relationship between the neurons expressed CRF1 receptor and the neurons secreting SP. In vivo submucosal plexus culture and RIA were combined to investigate the influence of CRF on SP secretion. Result All of the neurons expressed SP also expressed CRF1 receptor in submucosal plexus of guinea-pig. CRF could prompt the secretion of SP of submucosal plexus in vivo. Conclusion There is close relationship between CRF/CRF1 receptor and neurons expressed SP in enteric nervous system. It may suggest that CRF can prompt SP secretion through the CRF1 receptors expressed on neurons that originally expressed SP.
