摘要
目的:探讨正常及肾盂肾炎大鼠肾脏表面活性蛋白A(SP-A)的表达变化及黄芩甙对SP-A表达的影响。方法:将24只雄性Wistar大鼠随机分成4组,每组6只,A组:假手术+生理盐水;B组:假手术+黄芩甙;C组:模型+生理盐水;D组:模型+黄芩甙。用致肾盂肾炎大肠杆菌(uropathogenicE.coli)制备Wistar大鼠急性肾盂肾炎模型。采用免疫组化方法观察肾组织SP-A的表达,用逆转录聚合酶链反应(RT-PCR)方法观察SP-AmRNA的表达变化。结果:假手术组肾组织SP-A蛋白主要表达于肾小管上皮细胞,以外髓质处较为明显。模型组中SP-A表达的肾小管数量明显增多,染色变深,其阳性表达率于造模后显著高于假手术组,SP-AmRNA表达显著增强(P<0.05),模型+黄芩甙组中SP-A的表达较模型+生理盐水组增强(P<0.05)。结论:本实验证实了肾组织SP-A蛋白主要表达于小管上皮细胞,以外髓质处明显。急性肾盂肾炎模型大鼠的SP-A蛋白表达增高,SP-AmRNA表达增强。SP-A可能在急性肾盂肾炎的发病过程中起着重要的防御作用。
Objective To investigate the expression of surfaetant protein A (SP-A) in normal kidney and E. coil-induced phynonephritie kidney, and observe the effect of baiealin on SP-A expression. Methods Twenty-four male Wistar rats were randomly divided into four groups: sham plus NS group(group A); sham plus baiealin group(group B); pyelonephfitie plus NS group (group C); pyelonephfitie plus baiealin-treated group (group D) (n =6, each group) . Expression of SP-A in all kidney was measured by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) . Results SP-A labeling was observed in the renal tubular epithelial ceils in group A and B. In group C and group D, the expression of SP-A in tubular epithelial ceils significantly increased compared with that in group A and B. Expression of SP-A mRNA also increased significantly in group C and D ( P 〈 0. 05) . Conclusion After inducing pyelonephritis in rats with E. eoli, SP-A expression in mRNA level and protein level significantly increase, which indicates that SP-A may play an important role in host defence system.
