RP-HPLC测定扯根菜中槲皮素的含量

Determination of quercetin content in Penthorum chinese Pursh.by RP-HPLC

目的测定扯根菜中槲皮素的含量.方法采用RP-HPLC法, Aichrom Bond-AQ C18柱(250 mm×4.6 mm,5 μm),以甲醇-0.2%磷酸(56∶44)为流动相,流速1.1 ml·min-1,柱温35℃,检测波长370 nm.结果槲皮素进样量在0.0461～0.9220 μg范围内与峰面积呈很好的线性关系(r=0.9990),平均回收率为97.6%,RSD=1.86%.6批样品中槲皮素含量分别为2.16、2.25、2.21、2.26、2.08、2.05 mg·g-1.结论所建方法简便、准确、重复性好,可用于扯根菜中槲皮素的含量测定.

OBJECTIVE To establish a method to determine quercetin in Penthorum chinese Pursh.. METHODS An HPLC method was adopted. The column was Aichrom Bond- AQ C18 （250 mm × 4.6 mm, 5μm）. The mobile phase consisted of methanol - 0.2 % phosphoric acid （56：44）, with the flow rate of 1.1 ml·min^-1 and the UV detector at 370 nm. RESULTS The method displayed a good linearity for quercetin within the range of 0.0461 - 0.922 μg（ r = 0.9990）. The average recovery was 97.6%. The contents of six batches of sample were 2.16,2.25,2.21,2.26,2.08 and 2.05 mg·g^- 1 ,respectively. CONCLUSION This method is simple,accurate,reproducible and suitable for the determination of quercetin in Penthorum chinese Pursh..
