氨茶碱对K562细胞增殖与凋亡的影响

Effects of Aminophylline on Proliferation and Apoptosis in K562 Cells

目的为探讨磷酸二酯酶抑制剂在慢性粒细胞白血病细胞系增殖与凋亡中的作用,研究了非选择性磷酸二酯酶抑制剂氨茶碱对K562细胞系的影响。方法采用MTT检测观察细胞增殖变化,膜联蛋白V染色观察细胞凋亡率。结果MTT显示氨茶碱对K562细胞系增殖呈浓度依赖性抑制作用,膜联蛋白V染色显示氨茶碱诱导细胞凋亡率增加。结论氨茶碱可诱导K562细胞凋亡。

Objective To investigate whether and how phosphodiesterase （PDE） inhibitors modulate the proliferation, cell cycle and apoptosis in K562 ceils. The effects of aminophylline （AM） , a non- specific PDE inhibitor on K562 cells were explored. Mdthods MTT assay and annexin V staining were used to observe cell proliferation and apoptosis rate in AM - treated K562 cells. Results AM inhibited the growth of K562 cells in a concentration-dependent manner. FCM showed that AM intervention increased the fraction of annexin cells V^＋ cells. Conclusions AM may induce K562 cell apoptosis.
