联合转导人α1,3-半乳糖苷酶和α1,2-岩藻糖转移酶基因清除猪细胞表面异种抗原 被引量:6

摘要 猪细胞表面Galα1-3Galβ1-4GluNAc-R结构(称为αGal表位)是引起异种器官移植超急性排斥反应的主要原因,被称为主要异种抗原,由α1,3-半乳糖转移酶生成.α1,3-半乳糖苷酶特异性地水解αGal表位末端半乳糖,可清除这种主要异种抗原;α1,2-岩藻糖转移酶与α1,3-半乳糖转移酶竞争底物的作用也可使Galα1,3-Gal的生成量降低并阻止α1,3-半乳糖苷酶水解的产物重新生成Galα1,3-Gal,也具有清除主要异种抗原的作用.以猪胚胎成纤维细胞为靶细胞,联合转导人α1,3-半乳糖苷酶和α1,2-岩藻糖转移酶基因,获得的转基因细胞异种抗原清除率达84%,转基因细胞染色体数目和形态正常,为进一步获得低αGal抗原的体细胞克隆猪奠定了基础.
