人巨细胞病毒感染套式PCR基因诊断方法的研究

Genetic Diagnosis or Human Cytomegalovirus Infection by Nested Polymerase Chain Reaction

依据人巨细胞病毒主要早期基因核苷酸序列，建立了其套式ＰＣＲ诊断方法．并组装了较为实用的ＰＣＲ诊断试剂盒．该方法特异性强，敏感性较普通ＰＣＲ技术高２—３个数量级，标本处理方法简单，并可在６ｈ内判定结果，是一种在临床上值得推广应用的技术．

Upon the pulished nucleotide sequences of the major immediate-early gene of human cytomegalovirus,we established the PCR diagnosis methods including nested PCR method for detecting human cytomeglovirus,and then, packaged the practical PCR diagnosis kit.The nested-PCR of human cytomegalovirus was well specificially amplified the target DNA fragment, the sensitivity could increase 2～3 orders by nested-PCR to that of the PCR.The method of obtaining sample DNA was simple,and it was within 6 hours to get the analytical results. Therefore,the nested-PCR technology may be one method with higher applicational value.
