摘要
目的探讨中药醒脑启智胶囊对血管性痴呆小鼠脑组织海马细胞Bcl-2和Bax蛋白的影响。方法将120只昆明种小鼠随机分为假手术组、模型组、醒脑液高剂量组、醒脑液低剂量组、银杏叶液对照组、尼莫地平液对照组,每组20只,采用双侧颈总动脉结扎反复脑缺血再灌注的方法制备血管性痴呆小鼠模型。术后15d进行行为学实验和脑组织的病理形态学观察,并用流式细胞术检测各组海马细胞Bcl-2和Bax蛋白的含量。结果(1)行为学实验显示,模型组小鼠学习成绩与记忆成绩下降,各治疗组小鼠的学习成绩与记忆成绩均有提高(P<0·05)。(2)光镜下病理形态学显示,术后模型组小鼠脑组织海马区呈缺血性病理改变,各治疗组小鼠病变轻于模型组。(3)流式细胞术检测显示:模型组小鼠脑组织海马细胞Bcl-2和Bax蛋白含量高于假手术组(P<0·01),但二者的比值低于假手术组(P<0·01);各用药组与模型组比较,Bax蛋白降低(P<0·01),Bcl-2蛋白与Bcl-2/Bax的比值升高(P<0·05)。结论醒脑启智胶囊对血管性痴呆小鼠有明显治疗作用,其作用机制之一是调节海马细胞Bcl-2和Bax蛋白的表达。
Objective To investigate the effects of xingnaoqizhi capsule on Bcl - 2 and Bax proteins of hippocampus cell in experimental mice. Methods 120 Kunming mice were randomly divided into 6 groups: sham - operation group, model group, high -dose xingnaoqizhi group, low- dose xingnaoqizhi group, GinkgoBiloba control group, Nimodipine control group. Each group included 20 mice. We duplicated the model of vascular dementia mice with the operation of cerebral ischemic reperfusion. On the 15th day the morphologic changes of cerebral tissue were observed, and Bcl -2 as well as Bax proteins of the hippocampus cells in mice were determined by flow cytometry (FCM) after praxiology experiment. Results ( 1 ) The results of praxiology experiment showed that the learning and memorizing performance record of model group were failed and that of other groups were improved ( P 〈0. 05 or P 〈0.01 ). (2) The pathomorphology indicated that hippocampus of mice brain tissue took up the pathological changes of ischemia. The pathological change of each medical group was lighter than that of model group. ( 3 ) FCM showed that Bcl - 2 and Bax proteins of the hippocampus cells in mice of model group were higher than that of sham - operation group ( P 〈0. 01 ) , but Bcl -2/Bax was lower. At the same time, Bax protein of each medical group was lower, and Bcl-2 protein as well as Bcl - 2/Bax were higher than that of model group ( P 〈 0.05 or P 〈 0.01 ). Conclusion Xingnaoqizhi capsule could regulate Bcl - 2 and Bax proteins of the hippocampus cells to treat vascular dementia.
