摘要
目的:探讨免疫抑制剂普乐可复(FK506)和/或骁悉(MMF)联合抗包虫病药阿苯哒唑脂质体(L-ABZ)治疗泡球蚴感染大鼠心脏移植的疗效。方法:建立SD大鼠对Wistar大鼠心脏移植模型。泡球蚴感染的52只Wistar 雄性大鼠随机分为6组,分别单独或联合给予FK506、MMF、L-ABZ进行治疗,判断心脏排斥后,观察移植物存活时间。结果:FK506+MMF治疗组、感染对照组大鼠移植心脏存活时间分别为(16.11±8.04)d、(11.6±6.02)d,2组相比差异有统计学意义(P<0.05),其余各组大鼠移植心脏存活时间与感染对照组相比未见明显延长(P>0.05)。结论:(1)单独应用L-ABZ不会影响移植物的存活;(2)L-ABZ与FK506+MMF可能有一定的拮抗作用。
Objective: To study the effect of combined adminisitration of tacrolimus (FK506), mycophenolic acid (MMF), and L-ABZ on SD-to-Wistar rat cardiac graft survival. Methods: Fifty-two healthy SD rats and 52 E.m. infected Wistar rats were used in the study. The animals were randomly divided into: Groupl SD-to-Wistar(n = 8), without any treatment; Group 2 SD-to-Wistar(n = 9), L-ABZ 50 mg·kg^-1·d^-1 Group 3 SD-to-Wistar(n = 8), FK506 1 mg·kg^-1·d^-1; Group 4 SD-to-Wistar(n = 9), FK506 1 mg·kg^-1·d^-1、L-ABZ 50 mg·kg^-1·d^-1; Group 5SD-to-Wistar(n = 9), FK506 1 mg·kg^-1·d^-1、MMF20 mg·kg^-1·d-1; Group6 SD-to-Wistar(n = 9), FK506 1 mg·kg^-1·d^-1、MMF20 mg·kg^-1·d^-1、L-ABZ 50 mg·kg^-1·d^-1. All the agents were administrated orally one time per day. Graft samples were harvested for histopathological examination and immunohistochemical study. Results: FK506+MMF can significantly prolong the survival time of graft vs control (16.11±8.04 vs 11.6±6.02, P〈0.05). The survival time of the grafts in all other groups were not significantly prolonged. Conclution: (1) L-ABZ could not affect the survival of graft alone. (2) Use of L-ABZ might antagonize the effect of the FK506+MMF.
