摘要
目的观察人牙周膜成纤维细胞(HPDLF)受动态张应力作用后细胞培养液中基质金属蛋白酶-9(MMP-9)的合成与表达,探讨不同张应力对HPDLF分泌MMP-9的影响。方法对体外培养的第6代HPDLF在膜式动态张应力-体外细胞培养研究体系中进行不同应力水平、频动范围及时相点的应力加载,以双抗体夹心ABC-ELISA定量方法检测加力后细胞培养液分泌MMP-9的含量。结果HPDLF受0张应力时,MMP-9维持在一个较低水平;受到动态张应力作用后,MMP-9的分泌量有不同程度的增加,特别是动态张应力组1,细胞受到5kPa^0~5kPa(1kPa=7.50mm Hg)动态张应力作用后,MMP-9表达明显增加。结论不同张应力可引起MMP-9含量发生明显的变化,对临床上选择最适矫治力可能起到重要的作用。
Objective To investigate the amount of MMP-9 in human periodontal ligament fibroblast (HPDLF) under different dynamic tension stress in vitro. Methods HPDLF were cultivated artificially as the cells to be tested,and then they were loaded with the cellular mechanical research system of different frequency,magnitude and different times(16 h,24 h and 32 h). The level of MMP-9 was tested by use of quantitative ABC-ELISA assay. Then the relationship among tension stress , the time and the level of MMP-9 was detected. Results It was found that the amount of MMP-9 kept a low level following the 0 tension stress,and some HPDLF were taken place regular changes following the different dynamic tension stress groups, furthermore the changes appeared more obviously in dynamic tension stress group 1 (5 kPa- 0 -5 kPa). Conclusion Different tension stress patterns take different effects on the secretion of MMP-9. It may play a very important role in choosing the best orthodontic force in clinic.
