摘要 目的:探讨脑源性神经营养因子(BDNF)和胰岛素样生长因子1(IGF-1)对大鼠皮层神经干细胞向神经元定向分化的影响。方法:采用细胞培养技术、免疫细胞化学方法观察BDNF和IGF-1对皮层神经干细胞向神经元定向分化的影响。结果:皮层神经干细胞在去除丝裂原后开始进行分化,至7 d可分化为成熟神经元,神经元特异烯醇化酶(NSE)明显阳性。BDNF、IGF-1组与对照组相比,定向分化为神经元的比率增加21.7%(P<0.01)。结论:BDNF,IGF-1对皮层神经干细胞向神经元的定向分化具有促进作用。 Objective: To observe the effect of brain-derived neurotrophic factor (BDNF) and insulin-like growth factor-1 ( IGF-1 ) on the directional differentiation of cortex neural stem cells into neurons. Methods: The differentiation of neural stem cells into mature cholinergic neurons was observed by using cell culture and immunocytochemical method. Resulits:The stem cells isolated from the cerebral cortex were induced to differentiate after mitogen was removed. Mature neurons with positive reaction to neuronal specific enolase (NSE) were not observed until the 7th day. The proportion of m ture neurons was 21.7% higher in the coincubation of BDNF and IGF-1 than in the control group ( P 〈 0.01 ). Conclusion:BDNF and IGF-1 can promote the directional differentiation of cortex neural stem cells into neurons and increase the differential proportion.
