开胸冠状动脉结扎法制作大鼠心肌梗死模型 被引量:17

摘要 目的探索病理性大鼠心肌梗死模型的制作方法。方法在全身麻醉机械通气支持下,开胸结扎大鼠心脏左冠状动脉,同时记录手术前后心电图,并于术后1、2及3 d处死并取其心脏进行病理组织学检查。结果开胸结扎大鼠心脏左冠状动脉后,25只大鼠中 5只存活,7只死于心脏停搏,8只死于术中大出血,5只死于呼吸衰竭。病理组织学检查示存活的大鼠均出现左心室心肌大片断裂,心肌组织疏松,溶解,并可见大量炎性细胞浸润,纤维组织增生;或呈典型的心肌凝固性坏死病理表现。所有大鼠均出现心律失常,室性停搏或心室纤维颤动。结论通过开胸冠状动脉结扎法制作大鼠心肌梗死模型对大鼠的创伤大,成功率较低(20%)。操作技术的熟练程度、良好稳定的通气是手术成功的关键。 Objective To investigate the method how to make rat myocardial infarction model. Methods Keeping ventilation by using general anesthesia machinery, and performing left thoracotomy, then ligated left eoronary artery of the rat's heart. Recording the electrocardiogram before surgery., and taking rat's heart to carry on the histopathologic examinations on day 1, 2 and 3 after the surgery. Results After receiving the surgery, as above, 5 of 25 rats survived, 7 rats died of asysrole, 8 rats died of serious hemorrhage, 5 rats died of the respiratol7 failure. The histopathologie examination of the survival rats showed the myocardium of the left ventricle had big fragmentation. The cardic muscle appeared looseness, dissolution, accompanying with the mssive inflammation cell infiltration and fiber tissur proliferation; or some showed typical cardie mutie coagulation necrosis. Arrhythmia, ventricular arrest or ventricular fibrillation were observed in all of rats. Conclusion Using thoracotomy and ligationg the left coronary artery as the method to make rat myocardial infarction model causes big injury to rats and the successful possibility is lower (20%). Skilled operation and keeping infarction stalbe ventilation are key factors to the success of the surgery.
