西洋参悬浮细胞系的建立及其生长特性的研究 被引量:11

摘要 通过对西洋参根外植体愈伤组织诱导和培养,筛选出了质地疏松、生长快的愈伤组织无性系;将该愈伤组织接种在液体培养基中进行振荡培养,经多次传代、筛选获得了分散程度好的高产西洋参悬浮细胞系12-1。在此基础上,建立并优化了西洋参悬浮细胞培养工艺体系:温度(24±1)℃,pH值6.0,生长周期是25d,培养4d后再换液继代一次,培养20d后补料。细胞悬浮培养结束后,细胞的生长量及人参皂甙含量分别达到23.65g/L和677.25mg/L。 Light yellow translucent callus of incompact, rapidly grown, easily decentralized and subculture suited for a longterms and the optimum medium have been selectected by culture of explant from Panax quinquefolium root and culture of callus. A P. quinquefolium cell line was obtained when these callus were transferred to liquid medium of oscillation culture. On basis of this, the cell suspension culture system in P. quinquefolium was established and optimized including temperature (24 4±1)℃ , pH 6.0, and growth cycle 25 days. The feeding method was established. The dry weight of cell suspension culture and the saponin yield were 23.65g/L and 677.25mg/L.
