利用正交设计优化产氨基酰化酶的米曲霉的培养条件

Optimization of Culture Conditions of Aspergillus oryzae 3042 Producing L-aminoacylase with an Orthogonal Experimental Design

摘要 采用正交实验设计L9(34)方法,利用摇瓶培养研究了产氨基酰化酶的米曲霉的培养温度、培养基pH和培养时间3个因素对菌体酶活的影响。结果表明,在培养温度29℃,培养基起始pH 6.5以及培养时间50 h,其酶活达595 U/g以上。采用正交试验设计可以大大减小工作量,是一种高效率、快速、经济的实验设计方法。 The effect of culture conditions including culture temperature, medium pH value and culture time on culturing Aspergillus oryzae 3042 producing L - aminoacylase which is used for resolution of DL - methionine was studied in the shaking tlnsk. Based on the orthogonal experimental design L9(34), the three factors including medium pH, culture temperature and culture time which have respectively three levels were optimized. The results showed that under the optimal culture conditions of medium pH 6.5, culture temperature 29 ℃ and culture time 50 h, a high L - amlnoacylaso activity over 595 U/g was obtained. The orthognnal experimental design is a quick and efficient method to design a test and obviously reduce the work burden.
