摘要
目的研究西乐葆对A549细胞COX-2表达的影响。方法分别使用5、10和15μmol/mL的西乐葆对IL-1β诱导下A549细胞COX-2的表达进行干预,收集上清-20℃冻存用于检测PGE2,细胞用PBS洗两次提取RNA。不加西乐葆为对照组。实验重复4次。结果5、10和15μmol/mL的西乐葆干预组COX-2OD值/β-actinOD值分别为(1.4362±0.1061),(1.0142±0.079),(0.6896±0.097)与空白组(1.8058±0.132)相比较有明显降低,差异有显著性(P<0.05)。5μmol/mL组分别与10μmol/mL、15μmol/mL组相比,10μmol/mL组与15μmol/mL组相比都有统计学意义(P<0.05);5、10和15μmol/mL的西乐葆干预组PGE2分别为(17.25±0.98)×10-6mg/L,(11.33±1.49)×10-6mg/L和(10.23±2.00)×10-6mg/L较空白对照组(22.36±1.49)×10-6mg/L明显降低,差异有显著性(P<0.05)。5μmol/mL组分别与10μmol/mL、15μmol/mL组相比,10μmol/mL组与15μmol/mL组相比都有统计学意义(P<0.05)。结论西乐葆抑制人肺上皮细胞COX-2表达。
[Objective] This study reports whether a selective cox-2 inhibitor Celecoxib could inhibit the expression of cox-2 in A549 cell. [Methods] A549 cells were cultured in vitro with 5 ng/ml IL-1β and exposed to different concentrations Celecoxib. COX-2 mRNA was determined by RT-PCR. PGE2 was measured by ELISA. [Resuits] This study showed that different concentration Celecoxib suppress the expression of COX-2 in A549 cell and showed greater suppression at higher concentrations. [Conclusion] In this study, we demonstrate that a selective cox-2 inhibitor Celecoxib significantly suppress the expression of COX-2 in A549 cell and the effective suppress of gene is depend on the concentrations of Celecoxib.
