摘要 目的观察游泳运动对2型糖尿病大鼠肾周脂肪脂联素和骨骼肌GLUT4基因表达的影响。方法采用小剂量链脲佐菌素加高脂高热量饲料喂养的方法建立Wistar大鼠2型糖尿病模型,给予游泳运动训练8周,检测大鼠肾周脂肪脂联素mRNA和骨骼肌GLUT4mRNA表达的变化以及血糖、胰岛素、血脂等代谢指标。结果糖尿病模型组肾周脂肪脂联素mRNA和骨骼肌GLUT4mRNA表达显著降低,分别较正常对照组降低了45%(P<0.01)和43%(P<0.01),空腹血清胰岛素,甘油三酯、总胆固醇和游离脂肪酸水平较对照组显著升高;游泳运动组脂联素和骨骼肌GLUT4mRNA的表达较模型组显著升高,分别为模型组的1.39倍(P<0.05)和1.62倍(P<0.01),接近对照水平,空腹胰岛素较模型组显著降低(P<0.01),甘油三酯、总胆固醇和游离脂肪酸水平均显著低于模型组(P<0.05或P<0.01)。结论游泳运动可能通过提高脂肪脂联素和骨骼肌GLUT4mRNA的表达,改善胰岛素抵抗。 [Objective] To explore the effect of swim training on the expression of adiponectin and GLUT4 mRNA in type 2 diabetic rats. [Methods] Swim training was performed for 8 weeks in the type 2 diabetic Wistar rat model, which was induced by the intraperitoneal injection of a small dose of streptozotocin plus high fat high caloric laboratory chow. The expression of adiponectin mRNA in perirenal fat and GLUT4 mRNA in skeletal muscles were assed by RT-PCR and the change of blood glucose, serum insulin, blood lipid were measured. [Result] Compared with the control group, the expression of adiponectin mRNA and GLUT4 mRNA were decreased by 45% (P 〈0.01), 43% (P 〈0.01 ) respectively,fast serum insulin, serum triglyceride, total cholesterol and FFA were increased significandy in the model group; The expression of adiponectin mRNA and GLUT4 mRNA were increased by 1.39 times and 1.62 times in swimming group as compared with the control group(P 〈0.05 and P 〈0.01, respectively). Compared with the control group, fast serum insulin levels was decreased obviously (P〈0.01) and serum triglyceride, total cholesterol and FFA were decreased significantly in the training group (P 〈0.01 or P 〈0.05). [ Conclusion] Exercise can improve insulin resistance through increasing the expression of adiponectin mRNA and GLUT4 mRNA in type 2 diabetic rats.
