海南香蕉束顶病和花叶病的PCR检测

Detection of Banana Bunchy Top Virus and Cucumber Mosaic Virus in Hainan by PCR

用CTAB法从感染束顶病(BBTV)的海南香蕉叶片组织中提取DNA,以此为模板分别用特异引物对BBTV的组分Ⅰ、Ⅲ和Ⅳ进行扩增并测序,结果得到大小分别为607、490和326bp的特异片断;用CTAB法从感染花叶病(CMV)的病叶组织中提取RNA,反转录成cDNA,以此为模板用特异引物进行扩增测序,结果得到大小为557bp的特异片断。用PCR方法可从相当于100ng的叶片组织中检测到BBTV和CMV。

olymerase chain reaction （PCR） assay or reverse transcription （RT） PCR assay was used for the detection of banana bunchy top virus （BBTV） or cucumber mosaic virus （CMV） in Hainan respectively. 607 bp, 490 bp and 326 bp fragments of nucleotide sequence of BBTV component I , III and IV respectively, and 557 bp nucleotide fragments of coding sequences of the CMV coat protein were obtained. Using PCR method, BBTV or CMV can be detected from the banana leaf tissue equivalent to 100 ng.
