摘要 目的利用家兔左肺灌注模型,探讨三磷酸腺苷-氯化镁(ATP-MgC l2)对缺氧再灌注肺线粒体内钙离子浓度和肺组织水分的影响。方法观察ATP-MgC l2对再灌注肺组织线粒体内钙离子浓度和肺组织水分的影响。结果(1)家兔左肺灌注模型均获成功。(2)缺氧再灌注组线粒体内钙离子浓度显著高于正常对照和缺氧组(P<0.01);ATP-MgC l2加用再灌注组线粒体内钙离子浓度显著低于缺氧再灌注组(P<0.01)。(3)缺氧再灌注组肺组织湿干重比值(LW/D R)显著高于正常对照和缺氧组(P<0.01);ATP-MgC l2加用再灌注组LW/D R显著低于缺氧再灌注组(P<0.01)。结论本模型具有操作简便、成功率高的优点,是可行的肺离体研究模型;ATP-MgC l2明显抑制家兔缺氧再灌注肺线粒体内钙沉积和肺组织水肿,提示ATP-MgC l2对肺缺血再灌注损伤具有保护作用。 OBJECTIVE The purpose of this study is to establish reliable and appropriate rabbit left lung perfusion model and to clarify whether ATP - MgCl2 can influence calcium concentration in mitochondria during lung hypoxia/reperfusion injury in the model. METHODS First, we established rabbit single left lung perfusion model. Second, we use this model to clarify influence of ATP - MgCl2 on calcium concentration in mitochondria during lung reperfusion injury. RESULTS ( 1 ) Rabbit left lung perfusion model were successfully established in all 30 cases. (2) The free calcium concentration in mitochondria in hypoxia/reperfusion group was significantly higher than that in normal control and hypox- ia groups ( P 〈 0.01 ) ; In ATP - MgCl2 added reperfusion group was significantly lower than that in hypoxia/reperfusion group ( P 〈0.01 ) ; Lung wet/dry weight ratio in ATP - MgCl2 added reperfusion group was less than that in hypoxia and hypoxia/reperfusion groups ( P 〈 0.01 ). CONCLUSION The rabbit left lung perfusion model is a useful and reliable model for lung research. ATP - MgCl2 may have protective effect on the lung hypoxia/reperfusian injury through inhibition of mitochondrial calcium accumulation and reduction of lung tissue edema.
