摘要
目的:研究不同浓度三氧化二砷(As2O3)对人骨髓增生异常综合征难治性贫血伴原始细胞过多型(MDS-RAEB)细胞株MUTZ-1细胞增殖的影响,并探讨其可能的作用机制。方法:采用MTT法分析细胞生长曲线,用细胞形态学观察细胞凋亡。结果:低浓度As2O3(0.05~0.25μmol/L)对MUTZ-1细胞无明显生长抑制作用,高浓度As2O3(1.0~20.0μmol/L)对MUTZ-1细胞有明显生长抑制作用,且呈浓度、时间效应-依赖关系(P<0.01)。结论:低浓度As2O3(0.05~0.25μmol/L)对MUTZ-1细胞无明显生长抑制作用。高浓度As2O3(1.0~20.0μmol/L)对MUTZ-1细胞有明显生长抑制作用,且呈浓度、时间效应依赖关系。
Objective To investigate the inhibition effect of As2O3 on the growth of human MDS-RAEB cell line MUTZ-1 cells and to explore the possible cellular and molecular mechanisms. Method The experiment use the method of MTT to investigate the inhibition effect of arsenic trioxide (As2O3) on the growth of human MDS-RAEB cellline MUTZ-1 Cells and to explore the possible cellular and molecular mechanism. Results Low concentration of As2O3 (0.05-0.25μmol/L)had no marked growth inhibition effect on MUTZ-1 cells,high concentration of As2O3 (1.0-20.0μmol/L)had marked growth inhibition effect on MUTZ-1 cells.After 48h treatment with2.0μmol/L As2O3. Conclusion Low concentration of As2O3 (0.05-0.25μmol/L)had no marked growth inhibition effect on MUTZ-1 cells, high concentration of As2O3 (1.0-20.0μmol/L)had marked growth inhibition effect on MUTZ- 1 cells.
