慢性乙型肝炎患者血清对肾小管上皮细胞凋亡的影响 被引量:1

摘要 目的研究慢性乙型肝炎患者血清对体外培养的肾小管上皮细胞的影响。方法以体外培养的人近端肾小管上皮细胞(HK-2)与慢性乙型肝炎患者血清共同培养72 h后,用流式细胞仪检测HK-2凋亡率,以健康人血清作对照。分析转化生长因子β1(TGF-β1)与HK-2凋亡之间的关系。结果慢性乙型肝炎患者血清组HK-2凋亡率明显高于对照组(P<0.01),HBV-DNA阳性血清组HK-2凋亡率高于HBV-DNA阴性组(P<0.01),且HK-2凋亡比例与血清TGF-β1水平成正相关(r=0.562)。结论慢性乙型肝炎患者血清可诱导体外培养的肾小管上皮细胞凋亡,这可能与TGF-β1升高及病毒复制活跃程度有关。 Objective To investigate the effect of transforming growth factor-β1 (TGF-β1 ) in sera of chronic hepatitis B (CHB) ca- ses on apoptosis of renal tubular epithelial cells in vitro. Method The normal human kidney proximal tubular cells (HK-2) were cultured in the presence of sera of CHB, and the apoptosis of HK-2 were assayed by Flow Cytometer after 72 hours. The sera from healthy persons were as the control. The relationship between the levels of TGF-β1 , HBV-DNA in sera of CHB and the apoptosis of HK-2 were analyzed. Result The apoptosis rates of HK-2 incubated with CHB sera were significantly higher than those with the control sera (P 〈 0. 01 ). The apoptosis rates of HK-2 incubated with HBV-DNA positive sera were higher than those with HBV- DNA negative (P 〈 0. 01 ) and positively correlated with the TGF-β1 levels in sera (r = 0. 562). Conclusion The sera from patients with chronic hepatitis B could induce apoptosis of HK-2. The apoptosis might relate to the TGF-β1 levels in sera and the active degree of replication of HBV.
