汞致肾细胞凋亡的量效关系研究 被引量:1

摘要 目的探讨汞致肾细胞凋亡的剂量-效应关系。方法昆明种小鼠32只随机分成4组。对照组腹腔注射容量为0.2 ml/10 g体重的0.9%的氯化钠溶液,染汞组分别腹腔注射0.751、.5、3.0 mg/kg的氯化汞溶液。透射电镜观察凋亡细胞形态;免疫组化法检测bcl-2和bax凋亡相关基因表达;流式细胞仪进行凋亡细胞百分数分析。结果随着染毒剂量的增加,肾近曲小管细胞由染色质聚集成块,进展到核破碎释放出凋亡小体、凋亡小体被邻近细胞吞噬;肾凋亡细胞百分数呈逐渐上升的趋势,呈现一定的剂量-效应关系;bcl-2表达逐渐减弱,而bax表达逐渐增强。凋亡细胞百分数与不同染毒剂量组bax/bcl-2的比值呈明显的正相关(r=0.966,P=0.034)。 Objective To explore the dose-effect relation of renal apoptosis induced by mercury. Methods 32 mice were randomly divided into four groups. The mice of control were given intraperitoneal injections with 0.9% saline. The different mercuric chloride ( HgCl2 ) group was administrated respectively with 0. 75mg HgCl2/kg, 1.5 mg HgCl2/kg and 3. Omg HgCl2/kg. With Transmission electron microscope, to observe the renal cell apoptosis morphologic changes; With immunochemistry to detect apoptosis related gene expression of bcl-2 and bax. The percent of the apoptosis cell was analyzed by Flowcytometry. Results Along with the dose increasing, the damage of microvilli is more severely, the nuclei manifest the typical apoptosis character;The increased trend of the percent of the apoptosis cell was shown in different dose groups ; The expression of bcl-2 is decreased progressively and bax is increased accordingly, the percent of the apoptosis cell is positive correlation with the ratio of bax/bcl-2( r = 0.966, P = 0.034). Conclusion Renal cell apoptosis induced by mercury show definite dose-effect relation.
