Transient expression of eukaryotic expression plasmid CEA in vitro and its detection

目的:检测含癌胚抗原(CEA)的真核表达质粒pWACEA在体外的瞬时表达。方法:用脂质体包裹含CEA的真核表达质粒pWACEA转染MCF7细胞,采用原位杂交和免疫组化技术检测CEA的表达。结果:转染细胞原位杂交和免疫组化均为阳性;对照标本未见阳性信号。结论:真核表达质粒pWACEA在体外可表达CEA分子,为进一步建立CEA阳性动物肿瘤细胞模型及研究CEA阳性肿瘤的免疫防治提供了基础。

Objective： To detect expression of the eukaryotic expression plasmid pWA-CEA in vitro. Methods： MCF-7 cell line was transfected by this mammalian expression plasmid pWA-CEA using lipofectamine reagent. The expression of CEA molecule was detected by in situ hybridization and immunocytochemistry technique. Results ： The positive signals were found in MCF-7 cells, but no positive signals were exhibited in contrast specimens. Conclusion ： CEA molecule can be expressed in vitro after transfection, which lay a good foundation for constructing animal CEA positive tumor cell model and studying the prevention and therapy of CEA positive tumor.
