高原胸部火器伤后肺组织G蛋白的异常表达

摘要 目的探讨高原胸部火器伤早期G蛋白变化及其意义.方法 18只体重相近(10~15kg)的健康杂种犬,随机分成3组(每组6只):平原对照组、高原移居组和高原世居组(世居组为高原杂种犬).用3%戊巴比妥钠静脉麻醉(30mg/kg),插入气管插管.于海拔3700m处用0.44g钢珠以(350±50)m·s-1的初速度从右第6肋间射入,造成右胸贯通伤,伤后立即封闭伤口,安放胸腔闭式引流.伤后12h,取双侧肺组织,检测G蛋白三个亚型(Gi,Gs,Gq)的表达情况.结果伤后12h各组右肺(伤侧)的总表达量均明显增高,与左侧(健侧)比较其差异有显著意义.各组左肺之间差异无显著意义.移居组右肺Gi、Gs、Gq的表达增高,与对照组及世居组比较其差异有显著意义.结论高原胸部火器伤后G蛋白表达异常增高,以高原移居组最为明显,这可能是高原胸部火器伤后肺水肿发生的重要环节之一. Objective To study the changes of G-protein expression on chest firearm trauma at high altitude in dogs. Methods 18 mongrel dogs (10 to 15kg)were randomly divided into 3 groups:control group, high altitude immigrant group and high altitude native group,6 in each.Anesthetized dogs were shot in the fight chest with steel bails with an initial velocity of 350±50m/s at high altitude (3700m above sea level).The wounds were closed and the dogs were given thoracic closed drainage after injury. 12 hours after injury, the fight and left lungs were taken out for G-protein measurement. Results 12 hours after injury, there were increasing expressions of Gi,Gs,Gq in fight lungs than those in the left in each group,the immigrant group showed a significant increase than other groups. Conclusions There were increasing expressions of G-protein in each group, especially in immigrant group after injury ; these changes may be one of the important links of lung edema after firearm trauma at high altitude.
