摘要
为观察光动力疗法合并化疗的疗效及作用机制,体外培养舌鳞状细胞癌细胞株,分别加入小剂量平阳霉素、VCR、5-FU、CDDP和MC,孵育24小时,将细胞阻断于某一周期时相,经HpD加红光处理后,明显增加疗效,产生协同作用。相反,先HpD加激光后,再经化疗药物作用24小时,不产生协同作用。
An experimental study on the therapeutic effect of chemotherapy combined with photodynamic therapy on the squamous cell carcinoma cell line (SCCT) cells, a cell line of squamous cell carcinoma of the tongue, was carried out. Cells were incubated with small dose of chemotherapeutic agents, cisplatin, 5 FU, vincristine, mitomycin and pingyangmycin for 24 h at 37℃, respectively. It was shown that the cells were arrested predominantly at a certain phase of the cell cycle. The cultured cells were followed by photodynamic therapy using hematoporphyrin derivative and red light irradiation. The photodynamic cell killing effect was significantly potentiated synergistically by the action of chemotherapy. The cells arrested at S phase was most sensitive to such a treatment, and cells arrested at M phase were less sensitive than those in G1 or G2 phase. However, when the PDT treated cells were followed by chemotherapy, no synergistic effect was observed. It was assumed that the synergistic effect on PDT might be due to an enhanced HpD uptake caused by an altered membrane permeability after chemotherapy.
